Although growing evidence from both in vivo and in vitro studies indicates that the reversed pH gradient in cancer cells may represent a promising therapeutic target, current mainstream treatments for advanced cancer continue to rely primarily on chemotherapy and molecularly targeted therapies, with relatively few clinical approaches specifically focused on regulating cancer cell pH.
There are two main therapeutic approaches that target the acidic pH of the tumour micro environment. One is buffer therapy, in which alkalizing agents are administered to neutralize protons, and the other is the inhibition of proton efflux transporters expressed on the cancer cell membrane
